ABSTRACT

Electrophorus spp. generate high-voltage electric discharges for defense and hunting, and low-voltage electric discharges (as other Gymnotiformes) for electrolocation and communication. Despite intense interest in the unusual electrogenic and electroreceptive capacities of electric eels, the other sensory systems of Electrophorus spp. are relatively poorly known. Here we describe the ontogenetic development and organization of the cephalic lateral-line canals in the lowland electric eel, Electrophorus varii. Preserved specimens of larvae, juveniles, and adults were examined to describe the spatial distribution of the canals and pores. Ontogenetic shifts of the cephalic lateral line formation were observed for each canal and support a hypothesis of non-synchronized development. The morphogenesis of cephalic canals in larvae and juveniles begins just before the onset of exogenous feeding. In adults, the cephalic sensory canals are formed separately from the skull and overlay cranial and mandibular bones and muscles. This study provides the first detailed description of the development and organization of the cephalic lateral-line system in Electrophorus varii.(AU)


Electrophorus spp. geram descargas elétricas de alta voltagem, usadas para defesa e caça, e (como os demais Gymnotiformes) descargas elétricas de baixa voltagem para eletrolocalização e comunicação. Apesar do grande interesse nas capacidades eletrogênicas e eletrorreceptivas incomuns dos poraquês, os outros sistemas sensoriais de Electrophorus spp. são relativamente pouco conhecidos. Aqui nós descrevemos o desenvolvimento ontogenético e a organização dos canais da linha lateral cefálica no poraquê da planície Electrophorus varii. Exemplares preservados de larvas, juvenis e adultos foram analisados para descrever a distribuição espacial dos canais e poros. Variações ontogenéticas na formação da linha lateral cefálica foram observadas para cada canal, apresentando um desenvolvimento não-sincronizado. Variações ontogenéticas da formação da linha lateral cefálica foram observadas para cada canal e suportam uma hipótese de desenvolvimento assincrônico. A morfogênese dos canais cefálicos em larvas e juvenis precede o início da alimentação exógena. Nos adultos, os canais sensoriais cefálicos se formam separadamente do crânio e sobrepõem os ossos e músculos cranianos e mandibulares. Este estudo fornece a primeira descrição detalhada do desenvolvimento e organização do sistema da linha lateral cefálica de Electrophorus varii.(AU)

ABSTRACT

In this study, the composition of the diet and the feeding activity of Electrophorus varii were evaluated. The influence of ontogeny and seasonality in these feeding parameters was also examined. Fish were collected in the Curiaú River Basin, Amazon, Brazil, from March 2005 to February 2006, during the rainy (January-June) and dry (July-December) seasons. Diet composition was characterized based on the analysis of stomach contents and feeding dynamics was assessed based on the Stomach Fullness Index (IR) calculated using stomach weight. Stomach content and RI data were grouped into four-cm size classes (40-80, 80-120, 120-160, and 160-200) and two seasonal periods (rainy and dry). The influence of ontogeny and seasonality in the diet was investigated through PERMANOVA, and in the food dynamics through ANOVA. The analysis of stomach contents revealed that fish were the most consumed preys by electric eels, especially Callichthyidae and Cichlidae. Diet composition and RI values of electric eels were not influenced by ontogeny and seasonality. Electric eels are fish predators, regardless of size class and seasonal period.(AU)


Neste estudo foram avaliadas a composição da dieta e a atividade alimentar de Electrophorus varii. A influência da ontogenia e da sazonalidade nestes aspectos da alimentação dos poraquês também foi examinada. Os peixes foram coletados na Bacia do rio Curiaú, Amazônia, Brasil, no período de março de 2005 a fevereiro de 2006, abrangendo os períodos chuvoso (janeiro-junho) e o seco (julho-dezembro). A dieta foi avaliada por meio da análise dos conteúdos estomacais e a dinâmica alimentar por meio do Índice de Repleção Estomacal (RI) baseado nos dados de peso do estômago. Os dados do conteúdo estomacal e do RI foram agrupados em quatro classes de tamanho em cm (40-80, 80-120, 120-160 e 160-200) e dois períodos sazonais (chuvoso e seco). A influência da ontogenia e da sazonalidade na dieta foi investigada por meio da PERMANOVA, e na dinâmica alimentar por meio da ANOVA. A análise do conteúdo estomacal mostrou que os peixes foram as presas mais consumidas pelos poraquês, especialmente Callichthyidae e Cichlidae. A composição da dieta e os valores de RI dos poraquês não foram influenciadas pela ontogenia e pela sazonalidade. Os poraquês são predadores piscívoros, independente da classe de tamanho e do período sazonal.(AU)

ABSTRACT

As they spend most of their time buried in the substrate and are not a fishery resource, heterenchelyids are seldom seen. These eels are characterized by their greatly reduced eyes, which are covered by semi-transparent skin, the absence of a pectoral fin or lateral line, and no pores on the head or body. Pythonichthys sanguineus is a particularly poorly-known species, with only eight scientific records from Cuba, Puerto Rico, Colombia, Venezuela, Guyana, and Suriname. The present study is based on six adult specimens of P. sanguineus captured by vessels of the shrimp trawling fleet along the northern Brazilian coast, in the vicinity of the Amazon Reef. We provide meristic, morphometric and DNA barcoding data. These findings provide insights into the distribution of the species off the northern Brazilian coast and contribute to the discussion about the southern limit of the Greater Caribbean fauna. (AU)


Heterenchelídeos passam a maior parte do tempo enterrados no substrato e raramente são vistos, não representando recursos pesqueiros importantes. Essas enguias são caracterizadas por seus olhos muito reduzidos e cobertos por pele semi-transparente, ausência de nadadeiras peitorais ou linha lateral, e ausência de poros na cabeça ou no corpo. Pythonichthys sanguineus é uma espécie particularmente pouco conhecida, com apenas oito registros em coleção zoológica para Cuba, Porto Rico, Colômbia, Venezuela, Guiana e Suriname. O presente estudo é baseado em seis espécimes adultos de P. sanguineus capturados pela frota industrial de arrasto de camarão-rosa na costa norte do Brasil, ao largo dos Recifes da Amazônia. Apresentamos dados merísticos, morfométricos e de DNA barcoding dos exemplares examinados. O registro fornece informações importantes sobre a distribuição da espécie na costa norte do Brasil, e contribui para a discussão sobre o limite sul da fauna do Grande Caribe.(AU)

ABSTRACT

Anisakiasis is a zoonotic disease induced by anisakid nematodes, and endoscopic inspection is used for a diagnosis or remedy for it. Anisakis simplex, Anisakis physeteris, and Pseudoterranova decipiens had been reported to be the major species causing human infections, particularly, in Japan. However, in Korea, recent studies strongly suggested that Anisakis pegreffii is the major species of human infections. To support this suggestion, we collected anisakid larvae (n=20) from 20 human patients who were undergone gastrointestinal endoscopy at a health check-up center in Korea, and molecular identification was performed on the larvae using PCR-RFLP analysis and gene sequencing of rDNA ITS regions and mtDNA cox2. In addition, anisakid larvae (n=53) collected from the sea eel (Astroconger myriaster) were also examined for comparison with those extracted from humans. The results showed that all human samples (100%) were identified as A. pegreffii, whereas 90.7% of the samples from the sea eel were A. pegreffii with the remaining 9.3% being Hysterothylacium aduncum. Our study confirmed that A. pegreffii is the predominant species causing human anisakiasis in Korea, and this seems to be due to the predominance of this larval type in the fish (sea eels) popularly consumed by the Korean people. The possibility of human infection with H. aduncum in Korea is also suggested.

ABSTRACT

The European eel Anguilla anguilla is a highly important market species which is also increasingly in danger of extinction in Europe. One of the ways of protecting the species in the natural environment, while maintaining its market supply, is its aquaculture, e.g. due to much higher survival rate under controlled conditions than in the wild. However, this can be done only when an effective artificial reproduction biotechnique is developed. The aim of this study was to use an ultrasound device to determine the sex and to perform a biopsy of the gonads, which is a part of complete protocol of eel artificial reproduction. The findings indicate that ultrasonography is highly useful in determining the sex in migrating eel (100% of sureness) and in performing high-precision biopsy of the gonads. The present method gives the possibility to quick determine the maturity of the female gonads. The application of ultrasonography (USG) is totally safe for fish and a portable ultrasound device can be used in both the laboratory and in the field...


A Enguia-Européia Anguilla anguilla é uma espécie de grande importância comercial e que apresenta também grande risco de extinção na Europa. Uma das formas para a proteção das espécies em seu ambiente natural e de manter o seu suprimento no comércio é a aquicultura, por exemplo, devido à taxa de sobrevivência em condições controladas ser muito superior a existente em condições naturais. Contudo isto só pode ser obtido quando é desenvolvida uma biotécnica de reprodução artificial eficiente. O presente trabalho foi delineado para utilizar um dispositivo ultrassonográfico a fim de determinar o sexo e de possibilitar a realização de biópsias das gônadas, o que é parte do protocolo complete de reprodução artificial de enguias. As constatações indicaram que a ultrassonografia é muito útil para a determinação do sexo das enguias (100% de exatidão) e também é capaz de possibilitar a realização de uma biópsia das gônadas de alta precisão. O método descrito oferece possibilidades para a rápida determinação da maturidade das gônadas das fêmeas. A aplicação da ultrassonografia é totalmente segura para peixes e o dispositivo portátil de ultrassonografia pode ser utilizado tanto no laboratório como em condições de campo...

ABSTRACT

Anisakis simplex sensu stricto (s.s.), Anisakis pegreffii, Anisakis berlandi (=A. simplex sp. C), and Anisakis typica are the 4 major species of Anisakis type I larvae. In the Republic of Korea (Korea), A. pegreffii, A. berlandi, and A. typica larvae in fish hosts has seldom been documented. In this study, molecular analysis was performed on Anisakis larvae from the sea eels (Astroconger myriaster), the major source of human anisakiasis in Korea, collected from Tongyeong City, a southern coastal area of Korea. All 20 sea eels examined were infected with Anisakis type I larvae (160 larvae; 8 per fish). Their species were analyzed using PCR-RFLP patterns and nucleotide sequences of internal transcribed spacers (ITS1, 5.8 subunit gene, and ITS2) and mitochondrial cytochrome c oxidase 2 (cox2). Most (86.8%; 112/129) of the Anisakis type I larvae were A. pegreffii, and 7.8% (10/129) were A. typica. The remaining 5.4% (7/129) was not identified. Thus, A. pegreffii is the major species of anisakid larvae in sea eels of the southern coast of Korea.

ABSTRACT

The study focused on the characteristics of circadian locomotor activity in the spiny eel, M. pancalus, kept under different photoperiodic conditions. Two experiments were conducted. Experiment 1 tested the light intensity dependent effect on circadian rhythmicity of the locomotor activity in spiny eel. Three groups of fish were entrained to 12L:12D conditions for 10 days. Thereafter, they were released to constant conditions for 15 days as indicated below: group 1-DD (0 lux), group 2- LLdim (~1 lux) and group 3-LLbright (~500 lux). The locomotor activity of the fish, housed singly in an aquarium, was recorded continuously with infrared sensors connected to a computer. More than 90% activity of the eels was confined to the dark hours suggesting nocturnal habit. Under constant conditions, the activity in 7/9 fish in group 1, 4/8 in group 2 and 3/8 in group 3, started free running with a mean circadian period of 24.48 ± 0.17 h, 23.21 ± 0.47 h and 25.54 ± 1.13 h in respective groups. Remaining fish in each group became arrhythmic. This suggests that spiny eel can be synchronised to LD cycle and under constant conditions they free run with a circadian period. However, their activity under LL is light intensity dependent; higher the intensity, more disruption in circadian locomotor activity. Experiment 2 was conducted to study the effect of decreasing night length (increasing photoperiod) on circadian locomotor activity. The fish were sequentially exposed to 16D (8L:16D), 12D (12L:12D), 8D (16L:8D), 4D (20L:4D) and 2D (22L:2D) for 10 days in each condition, thereafter, they were released in constant dark (DD= 0lux). The results showed that the duration of night length affects both, the amplitude and duration of locomotor activity. It can be concluded that the spiny eels are nocturnal and that their locomotor activity is under the circadian control and may be influenced by the photoperiod.

ABSTRACT

Heliconema hainanensis sp. nov. collected from Uroconger lepturus (Richardson) (Anguilliformes: Congridae), Muraenesox cinereus (Forsskål) and Congresox talabonoides (Bleeker) (Anguilliformes: Muraenesocidae) in the South China Sea was described using light and scanning electron microscopy. The new species differs from its congeners by the following morphology: pseudolabia, the number and arrangement of caudal papillae (4 pairs of pedunculate precloacal papillae arranged in 2 groups of 2 and 2 pairs and 6 pairs of pedunculate postcloacal papillae arranged in 4 groups of 1, 2, 1 and 2 pairs), the length of spicules [left spicule 0.51-0.69 mm, right spicule 0.20-0.27 mm, spicule (right:left) ratio 1:2.20-2.69] and the morphology of the female tail tip. In addition, specimens of the new species collected from the three different hosts and specimens of an unidentified species of Heliconema collected from U. lepturus were characterised using molecular methods by sequencing the internal transcribed spacer (ITS) of ribosomal DNA. Analyses and comparison of the ITS sequence of H. hainanensis sp. nov. with Heliconema sp. support the validity of the new species based on morphological observations. An identification key to the species of Heliconema is also provided.

ABSTRACT

There is a limitation to estimate vitamin B12 intake due to a lack of data on vitamin B12 content in many Korean foods. In this study, vitamin B12 content was determined in some seaweeds, fish, and shellfish and their product that are consumed in Korea using a modified microbioassay with Lactobacillus delbruecki ATCC 7830. Dried laver and dried seasoned and toasted laver contained very high levels of vitamin B12 (66.8 and 55.2-71.3 microg/100 g, respectively. Sea lettuce and seaweed fulvescene also contained high vitamin B12 content of 5.47-9.41 and 6.46-7.20 microg/100 g, respectively, whereas sea mustard and sea tangle contained low levels of vitamin B12; vitamin B12 was not detected in seaweed fusifome. Pacific saury, trout, sea-bass, or squid contained 12.01, 2.00, 0.49 and 2.33 microg vitamin B12/100 g, respectively. Ochellatus octopus, and naked sand lance contained 0.72-1.43 and 3.68 microg vitamin B12/100 g, respectively. Dried Alaska pollack contained 0.19-2.64 microg vitamin B12/100 g. Shellfish such as little neck clam and small ark shellfish contained high levels of vitamin B12 of 30.5-40.5 microg/100 g, and mussel and abalone contained 17.71 and 7.82 microg/100 g, respectively. Of unique Korean traditional fermented seafood products, salt-fermented products of squid (2.91 microg/100 g), clams (34.31 microg/100 g), Alaska pollack roe (9.98-12.02 microg/100 g), hairtail guts (4.58 microg/100 g) or small shrimp (0.58-1.55 microg/100 g), and fish sauce from anchovies (1.52-1.78 microg/100 mL), sand eel (0.22-0.24 microg/100 mL) or small shrimp (0.19-0.78 microg/100 mL) were analyzed. A few commercial brands of flying fish roe (0.73-1.73 microg/100 g), canned tuna (0.40 microg/100 g), and fried fish paste (0.25-0.69 microg /100 g) were also analyzed. In conclusion, vitamin B12 content in these foods, chosen considering the Korean food culture, should contribute to improve the present vitamin B12 food database. It may be helpful to estimate vitamin B12 intake more correctly than before, and provide additional information for dietary education related to vitamin B12 and meal management.

ABSTRACT

The speckeld worm eel Myrophis punctatus lives in high-densities assemblages, and usually digs through, or lies on the substrate. These behaviours could lead to chemical marks on the substrate and could modulate the spatial distribution in this species. We tested the hypothesis that the spatial choice of the speckled worm eel is modulated by the presence of conspecific odour on the substrate. Here, we showed that the speckled worm eel avoids the substrate area containing the conspecific odour, indicating that this chemical cue modulates the eel's spatial decision. The eels clearly detected the conspecific's odour. This perception might indicate the presence of conspecifics into the substrate. Since the eels avoided an area containing conspecific odour, we suggest this may be a response that avoids the consequences of invading a resident-animal's territory.


A enguia mirongo-mirim Myrophis punctatus vive em agrupamentos de alta densidade populacional e comumente se enterra ou permanece sob o substrato. Esses comportamentos podem levar a marcas químicas no subtrato e podem, portanto, modular o uso do espaço nessa espécie. Neste estudo, testamos a hipótese de que a preferência espacial da enguia mirongo-mirim é influenciada pela presença de odor do animal coespecífico no subtrato. Mostramos que as enguias evitam a área que contém tal odor, indicando que as decisões de ocupação espacial podem ser influenciadas por pistas químicas de coespecíficos. As enguias claramente detectaram o odor de um animal coespecífico e essa percepção poderia ser um indicativo da presença de um coespecífico enterrado no substrato. Visto que elas evitam uma área contendo tal odor, sugerimos que isso poderia ser uma resposta para evitar invadir o território de um animal residente.

ABSTRACT

This paper described aspects of feeding behavior of Myrichthys ocellatus in the Western Atlantic, using diving observations and additional data from literature. The feeding behavior of M. ocellatus was characterized by scanning the sand surface and searching for buried preys, using its head and tail as a searching tool. Twelve species of reef fish were detected as followers of M. ocellatus, including three new records of follower species. The food overlap index between M. ocellatus and most follower species was low.(AU)


O presente estudo descreveu aspectos do comportamento alimentar de Myrichthys ocellatus no Atlântico ocidental, através de observações subaquáticas, complementadas com dados compilados da literatura. Myrichthys ocellatus caracteriza-se por buscar presas entocadas ou enterradas no substrato não consolidados e por usar a cauda e a cabeça como ferramentas de busca. Doze espécies de peixes recifais foram observadas como seguidoras de M. ocellatus, incluindo três novas ocorrências. O índice de sobreposição alimentar entre M. ocellatus e a maioria das espécies seguidoras foi baixo.(AU)

ABSTRACT

The order Anguilliformes comprises 15 families, 141 genera and 791 fish species. Eight families had at least one karyotyped species, with a prevalence of 2n = 38 chromosomes and high fundamental numbers (FN). The only exception to this pattern is the family Muraenidae, in which the eight species analyzed presented 2n = 42 chromosomes. Despite of the large number of Anguilliformes species, karyotypic reports are available for only a few representatives. In the present work, a species of Ophichthidae, Myrichthys ocellatus (2n = 38; 8m+14sm+10st+6a; FN = 70) and four species of Muraenidae, Enchelycore nigricans (2n = 42; 6m+8sm+12st+16a; FN = 68), Gymnothorax miliaris (2n = 42; 14m+18sm+10st; FN = 84), G. vicinus (2n = 42; 8m+6sm+28a; FN = 56) and Muraena pavonina (2n = 42; 6m+4sm+32a; FN = 52), collected along the Northeastern coast of Brazil and around the St Peter and St Paul Archipelago were analyzed. Typical large metacentric chromosomes were observed in all species. Conspicuous polymorphic heterochromatic regions were observed at the centromeres of most chromosomes and at single ribosomal sites. The data obtained for Ophichthidae corroborate the hypothesis of a karyotypic diversification mainly due to pericentric inversions and Robertsonian rearrangements, while the identification of constant chromosome numbers in Muraenidae (2n = 42) suggests a karyotype diversification through pericentric inversions and heterochromatin processes.

ABSTRACT

This study was conducted to investigate food and meal preference of workers (435 male and 212 female) in the Chonnam Yeosu industrial area. The results of the survey were analyzed by principal components analysis. The results were obtained as follows: 19.3% of the subjects were twenties, 28.3% were thirties, 28.8% were forties and 23.7% were fifties. Females liked rice rolled in dried laver, rice cakes, janchi-noodle and breads, on the other hand males liked thick beef soup, loach soup, an eel stew and soju. The young liked instant noodles, fried chicken, sweet and sour pork, pork cutlet, pizza, hamburger, ham, sausage and fruit beverage, on the other hand the old liked a fish pot stew, loach soup, eel stew, fish boiled in soy with spices, panfried fish, sea slug, ascidian, bunder, green laver and boiled burdock-lotus root in soy. Females who are young liked hamburger and sweet and sour pork whereas males who are young liked instant noodles, pork cutlet, ham, sausage and fruit beverage. Aged females liked fish boiled in soy with spices, panfried fish, bunder, green laver, boiled burdock-lotus root in soy whereas aged males liked a fish pot stew, loach soup, eel stew, sea mussel and oysters. Boiled rice was located within the middle irrespective of age and sex, but noodles, wheat flour meal, fast foods and fruits were situated at the young female side. Aged males liked soup and pot stew. Young males liked meats and eggs whereas fish and shellfish and kimch were located at the aged people side. Aged females liked sea weeds and most people disliked vegetables but females liked some vegetables irrespective of age. Processed foods, salted foods, and alcohol were generally disliked foods by subjects but males liked those foods. Soybean curd was liked more males than females, and teas, except coffee, was liked by males.

ABSTRACT

Despite the progresses done in the field of teleostean phylogeny in the last decades, recent studies continué to raise questions concerning the higher-level relationships of this remarkably diverse group of fishes. The main aim of the present work is to help to clarify teleostean higher-level relationships. For that purpose, we undertook a cladistic analysis including 70 terminal taxa of 20 different orders and 271 morphological characters, concerning mainly osteological and myological structures of the cephalic region , pectoral girdle and fins and anterior vertebrae. In the consensus cladogram obtained, the elopomorphs appear as the most basal extant teleosts. The osteoglossomorphs included in the analysis are grouped in a monophyletic clade, which is the sister-group of the remaining non-elopomorph teleosts. The Otocephala, the Clupeiformes, and the Ostariophysi appear as monophyletic clades, thus contradicting the results of some recent molecular cladistic analyses placing the Alepocephaloidea inside the Otocephala. In fact, the monophyly of the Argentiniformes (Alepocephaloidea + Argentinoidea) is well supported by the cladistic analysis of the present work. This cladistic analysis also provides support for the monophyly of the Alepocephaloidea, of the Argentinoidea, of the Galaxioidea + Osmeroidea, and of the Esociforines. However, it does notprovide strong evidence to resolve the relationships between the Argentiniformes, Salmoniformes, Esociformes, Osmeriformes and Neoteleostei, although it does indicate that the salmoniforms might be closely related to the Neoteleostei and that the Esociformes and the Osmeriformes might constitute a monophyletic unit. The monophyly of the Cypriniformes + Characiformes + Gymnotiformes + Siluriformes, of the Characiformes + Gymnotiformes + Siluriformes and of the Gymnotiformes + Siluriformes is well supported.


A pesar de los avances realizados en relacion a la filogenia de los teleosteos en las últimas décadas, los estudios recientes siguen planteando cuestiones relativas a los altos niveles de relacion de este notable grupo de diversos peces. El principal objetivo del presente trabajo es contribuir a aclarar los altos niveles de relacion de teleosteos. Con este propósito, se llevó a cabo un análisis cladístico entre 70 taxones terminales de 20 órdenes diferentes y 271 caracteres morfológicos, principalmente en relacion con estructuras osteológicas y miológicas de la region cefálica, cintura escapular y las aletas anteriores y vértebras. En el cladograma de consenso obtenido, los elopomorfos aparecen como los teleosteos más básicos existentes. Los osteoglosomorfos incluidos en el análisis se agrupan en un ciado monofilético, que es el grupo hermano de los restantes teleosteos no elopomorfos. Los Otocephala, los clupeiforines, y los ostariofisios aparecen como ciados monofiléticos, contradiciendo así los resultados de algunos análisis moleculares cladísticos recientes incluyendo los Alepocephalidae dentro Otocephala. De hecho, la monofilia de los Argentiniformes (Alepocephaloidea + Argentinoidea) está bien apoyada por el análisis cladístico del presente trabajo. Este análisis cladístico también proporciona apoyo para la monofilia de los Alepocephaloidea, de los Argentinoidea, de los Galaxioidea + Osmeroidea, y de los Esociformes. Sin embargo, no proporciona pruebas sólidas para resolver las relaciones entre los Argentiniformes, Salmoniformes, Esociformes, Osmeriformes y Neoteleostei, aunque indica que los salmoniformes podrían estar estrechamente relacionados con los Neoteleostei, y que los Esociformes y los Osmeriformes podrían constituir una unidad monofilética. La monofilia de los Cypriniformes + Characiformes + Gymnotiformes + Siluriformes, de los Characiformes + Gymnotiformes + Siluriformes y de los Gymnotiformes + Siluriformes está bien apoyada.

ABSTRACT

The pharmacotherapyfor Alzheimer's disease (AD) includes the use of acetylcholinesterase inhibitors (AChEI). Recent investigations for novel AD therapeutic agents from plants suggested that Tabernaemontana genus is a promising source of novel anticholinesterasic indole alkaloids. In this work two fast screening techniques were combined in order to easily identify novel cholinesterase inhibitors (ChEI). Gas chromatography-mass spectrometry (GC-MS) of the less polar alkaloidic fractions obtained from the acid-base extraction of the stalk of T. laeta revealed thirteen monoindole alkaloids, four of them confirmed by co-injection with previously isolated alkaloids. The others were tentatively identified by mass fragmentation analysis. By gas chromatography with flame ionization detection (GC-FID) and using isatin as internal standard, affinisine and voachalotine were determined as major compounds. These fractions and fourteen previously isolated alkaloids, obtained from root bark of T. laeta and T. hystrix were investigated for acetyl (AChE) and butyrylcholinesterase (BuChE) inhibitory activities by the modified Ellman's method in thin layer chromatography(TLC-ChEI). Results showed selective inhibition of the alkaloids heyneanine and Nb-methylvoachalotine for BuChE, and 19-epi-isovoacristine for AChE, whereas olivacine, affinisine, ibogamine, affinine, conodurine and hystrixnineinhibited both enzymes. In addition to confirming that monoterpenoid indole alkaloids can be novel therapeutic agents for AD, this is the first report of the ChEI activity of olivacine, a pyridocarbazole alkaloid.


Dentre os tratamentos da doença de Alzheimer (DA) está o uso de inibidores da enzima acetilcolinesterase. Pesquisas recentes visando a descoberta de novos agentes terapêuticos naturais para esta doença sugerem que o gênero Tabernaemontana é uma fonte promissora de alcalóides indólicos anticolinesterásicos. Neste trabalho, duas técnicas de análise em mistura foram associadas de modo a identificar facilmente novos inibidores colinesterásicos. A cromatografia em fase gasosa acoplada a espectrometria de massas (CG-EM) das frações alcaloídicas apolares, obtidas da extração ácido-base do caule de T. laeta, revelou a presença de treze alcalóides monoindólicos, quatro deles confirmados por co-injeção com padrões previamente isolados. Os outros alcalóides foram tentativamente identificados pelo padrão de fragmentação de massas. Por cromatografia em fase gasosa com detecção por ionização de chama (CG-DIC) e utilizando isatina como padrão interno, affinisina e voachalotina foram identificadas como substâncias majoritárias. As frações alcaloídicas obtidas e os quatorze alcalóides previamente isolados das raízes de T. laeta e T. hystrix foram analisados quanto à atividade inibitória das enzimas acetil (AChE) e butirilcolinesterase (BuChE) pelo método de Ellman em cromatografia em camada delgada (CCD-ChEI). Os resultados revelaram uma inibição seletiva dos alcalóides heyneanina e Nb-metilvoachalotina para BuChE e de 19-epi-isovoacristina para AChE, enquanto que olivacina, affinisina, ibogamina, affinina, conodurina e hystrixnina inibiram ambas as enzimas. Além de confirmar que alcalóides indólicos monoterpênicos são agentes terapêuticos promissores para o tratamento da DA, este é o primeiro relato da atividade anticolinesterásica de olivacina, um alcalóide piridocarbazólico.

ABSTRACT

Reproduction of the fish Gymnothorax equatorialis (Pisces: Muraenidae) in Jalisco and Colima, Mexico. A total of 707 Gymnothorax equatorialis were collected monthly in the Jalisco and Colima coast, Mexico, from December 1995 to December 1998 and from August to November 1999, in order to determine their reproduction patterns. Females outnumbered and had longer bodies (mean length 54.7 cm) than males (52.1 cm). The minimum, maximum and mean values of total fecundity were respectively 9 660, 99 992 and 32 029 eggs. The total body length at which 50 percent of individuals have ripe gonads (L50) was 43.7 cm for females and 42.7 cm TL for males. Ovaries had two main types of oocytes: small inmature in cromatin nucleolus phase (85.1 microm) and large mature in secundary vitellogenesis phase (701.6 microm). High numbers of spermatozoa were observed in the seminal tubes of ripe testicles, which presented a lobular development. Based in gonad features and temporal changes in the gonad-somatic index, it is concluded that G. equatorialis has two relatively short reproduction periods (at the middle and at the end of the year).


Con el fin de analizar los aspectos reproductivos de Gymnothorax equatorialis se recolectaron mensualmente 707 organismos en las costas de Jalisco y Colima, México, de diciembre de 1995 a diciembre de 1998 y de agosto a diciembre de 1999. Las hembras fueron más numerosas y presentaron una longitud (54.7 cm) mayor a la de los machos (52.1 cm). La fecundidad total mínima fue de 9 660 huevos, la máxima de 99 992 y la media fue 32 029 huevos. La talla en la que el 50 % de los individuos presentan gónadas maduras (L50) fue de 43.7 cm de longitud total en hembras y de 42.7 cm en machos. Los ovarios presentaron dos tipos de ovocitos, los pequeños inmaduros en fase cromatina nucleolo (85.1 μm) y los grandes maduros en fase de vitelogénesis secundaria (701.6 μm). En el testículo maduro se observó una gran cantidad de espermatozoides en el tubo seminal y el desarrollo del testículo es de tipo lobular. Con base en las características de las gónadas y la evolución temporal del índice gonadosomático, se concluye que G. equatorialis se reproduce dos veces al año (a mediados y finales del año).

BACKGROUND: The immunomodulatory effects of Korean mistletoe (Viscum album Coloratum) on the innate immune responses of eel (Anguilla japonica) were studied. METHODS: Mistletoe, Freund's complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control was injected into eel peritoneal cavities. RESULTS: Nitroblue tetrazolium (NBT)-positive cells in the head kidney of eel were significantly augmented by the second day post-injection of mistletoe. Reactive oxygen intermediates (ROI) were more produced in mistletoe-injected fish kidney leucocytes than in FCA-injected ones. The level of lysozyme activity in the serum of fish 2 days after injection with mistletoe was also significantly higher than that in the serum of the control fish. The optimal concentration of mistletoe in inducing the highest serum lysozyme activity was revealed to 500microgram/200 g of fish. In phagocytic activity assay, mistletoe-sensitized eel kidney phagocytes captured more zymosan than did the control fish. CONCLUSION: Korean mistletoe appeared to be a good activator of the non-specific immune responses of eel.

BACKGROUND: Vibrio vulnificus are divided into 3 biogroups based on their biochemical and serological properties and the existence of eel virulence. Only a few studies can be found on the biogroups of V. vulnificus in other countries, and no such studies have been done in Korea. OBJECTIVE: The aim of this study was to determine the presence of biogroups other than biogroup 1 among the V. vulnificus isolated from septic patients and from oysters in Korea. METHODS: A total of 103 isolates (53 from septic patients and 50 from oyster) were used. The API 20E system was used to confirm identification of the V. vulnificus. Conventional biochemical tests and vvA gene detection by polymerase chain reaction (PCR) were used to determine the biogroups. RESULTS: The clinical and oyster isolates showed results similar to the biochemical tests. All of the clinical and oyster isolates showed the biochemical pattern of biogroup 1. The vvhA gene was detected in all of the isolates. CONCLUSION: All of the V. vulnificus isolates from the septic patients and oysters in Korea belong to biogroup 1.

ABSTRACT

High throughput macromolecular structure determination is very essential in structural genomics as the available number of sequence information far exceeds the number of available 3D structures. ACORN, a freely available resource in the CCP4 suite of programs is a comprehensive and efficient program for phasing in the determination of protein structures, when atomic resolution data are available. ACORN with the automatic model-building program ARP/wARP and refinement program REFMAC is a suitable combination for the high throughput structural genomics. ACORN can also be run with secondary structural elements like helices and sheets as inputs with high resolution data. In situations, where ACORN phasing is not sufficient for building the protein model, the fragments (incomplete model/dummy atoms) can again be used as a starting input. Iterative ACORN is proved to work efficiently in the subsequent model building stages in congerin (PDB-ID: lis3) and catalase (PDB-ID: 1gwe) for which models are available.

ABSTRACT

This study was conducted to investigate the side dishes preference of Korean food with 403 college students (197 males and 206 females) in Seoul using questionnaires. Regarding preference of soups, it was shown that male students liked sollongt' ang and beef soup and female students liked soybean paste soup. Male students had higher preferences for sollongt' ang, beef soup, dried pollack soup, and kimchi soup than female students (p < 0.05). Both male and female students had the highest preferences for kimchi pot stew and soft-tofu pot stew, but the lowest preferences for fermented soybeans pot stew and frozen pollack pot stew (p < 0.05). Stewed beef with soy sauce was preferred the most by both of them but female students had lower preferences for stewed bean with soy sauce and stewed pepper with soy sauce than male students (p < 0.05). For the preference of panbroiled foods, stir-fried with marinated anchovy was disliked by both of them but stir-fried with marinated pork was shown to have high preference in male students and stir-fried with marinated kimchi was high in female students (p < 0.05). Besides, roasted ribs, cucumber salad, and korean cabbage kimchi were preferred the most by both of them. But male students had higher preferences for roasted eel, grilled todok, roots of broad bellflower salad, mustard leaf kimchi, and white cabbage kimchi than female students (p < 0.05).
